敲减BMI1对HeLa细胞增殖、侵袭、迁移及凋亡的影响Effects of knockdown of BMI1 on proliferation, invasion, migration and apoptosis of HeLa cell

摘要(Abstract):
目的探讨敲减BMI1对人宫颈癌HeLa细胞系增殖、迁移及凋亡的影响。方法 qRT-PCR方法分别检测宫颈癌及正常宫颈组织中BMI1的表达;用siBMI1及siNC转染人宫颈癌Hela细胞作为实验组及对照组,转染24h后,用CCK8实验、划痕实验、Transwell实验、流式细胞术分别检测敲减BMI1对Hela细胞增殖、侵袭、迁移、凋亡能力的影响。结果 qRT-PCR结果显示BMI1在宫颈癌组织中的表达显著高于正常宫颈组织;与对照组相比,CCK8实验结果显示敲减BMI1能够有效抑制宫颈癌HeLa细胞增殖的能力;划痕实验的结果表明敲减BMI1可抑制HeLa细胞的迁移能力;Transwell结果表明:敲减BMI1可抑制HeLa细胞的侵袭能力;流式细胞术结果证明:敲减BMI1可以促进HeLa细胞的凋亡。结论敲减BMI1能够显著抑制人宫颈癌HeLa细胞BMI1的表达,降低了HeLa细胞增殖、迁移、侵袭能力,并且可明确诱导HeLa细胞的凋亡。
