环磷酰胺对乳腺癌增殖、迁移和凋亡的影响及其机制Effect of Cyclophosphamide on proliferation, migration and apoptosis of breast cancer MCF-7 cells and its potential molecular mechanism

摘要(Abstract):
目的本文探讨环磷酰胺Cyclophosphamide对乳腺癌增殖、迁移和凋亡的影响及其机制研究。方法运用基因表达谱交互式分析(GEPIA)数据库分析乳腺癌组织中的基质金属蛋白酶9(mtrix metalloproteinases 9,MMP9)基因差异表达及预后情况。MTT法检测人乳腺癌细胞MCF-7细胞的增殖活性,TUNEL法检测细胞凋亡情况,划痕愈合实验检测MCF-7细胞水平迁移能力;反转录-聚合酶链反应PCR(RT-PCR)检测MMP9基因表达情况。结果 MMP9在乳腺癌中高表达,其基因表达量与乳腺癌患者总体生存率有关。CTX抑制乳腺癌细胞MCF-7的增殖、迁移,促进细胞凋亡,下调MMP9表达。结论 CTX能有效抑制乳腺癌MCF-7细胞增殖和迁移,诱导细胞凋亡,其机制可能与调控MMP9表达有关。
