截短型潜态相关肽对TGF-β诱导HSC-T6细胞增殖的影响Effect of Recombinant Truncated TGF-β Latent Associated Peptide on Proliferation of HSC-T6 cells

摘要(Abstract):
目的原核表达并纯化重组截短型TGF-β潜态相关肽(latency associated peptide,LAP),观察其对转化生长因子TGF-β(Transforming Growth Factor-β,TGF-β)诱导的大鼠肝星状细胞(Hepatic stellate cell,HSC)增殖的影响。方法 LAP各个片段重组蛋白小量培养经IPTG诱导表达,并通过Ni~(2+)亲和层析纯化,纯化产物进行SDS-PAGE电泳检测。通过MTT法检测浓度分别为2μg/mL、4μg/mL、8μg/mL、16μg/mL、32μg/mL、64μg/mL LAP-B对TGF-β诱导的HSC-T6细胞增殖作用的影响。结果成功诱导表达可溶性截短型重组蛋白LAP-B,大量培养后经镍柱亲和纯化获得目的蛋白,经SDS-PAGE检测为LAP截短型重组蛋白。MTT检测LAP-B在2~64μg/mL浓度范围内能抑制TGF-β诱导的HSC-T6细胞的增殖,并随着LAP-B蛋白浓度增加HSC-T6细胞增殖抑制越显著。结论成功诱导表达及纯化截短型重组蛋白LAP-B,LAP-B抑制了TGF-β诱导的HSC-T6细胞的增殖,为后续研究LAP的抗肝纤维作用提供实验基础。
