酵母双杂交技术筛选与STING蛋白相互作用的蛋白Screening for proteins interacting with protein kinase STING by yeast two hybrid system

摘要(Abstract):
目的应用酵母双杂交技术筛选出与干扰素刺激因子(stimulator of IFN genes,STING)相作用的分子,为深入研究STING蛋白的分子水平的功能奠定基础。方法利用基因克隆的方法构建酵母双杂交质粒pGBKT7-STING。应用酵母杂交技术从人胚肾cDNA库中筛选出与STING蛋白相互作用的蛋白,并在酵母中从新验证其相互作用。结果应用酵母双杂交系统筛选出与STING蛋白相作用的19个候选分子,成功构建pGBKT7-STING质粒,且可以在酵母中表达。结论应用酵母双杂交技术可筛选出与STING蛋白相互作用蛋白。
