MSCs-CM对人增生性瘢痕成纤维细胞的影响及其作用机制的研究Effect of MSCs-CM on human hypertrophic scar fibroblasts and its mechanism

摘要(Abstract):
目的研究骨髓间充质干细胞(mesenchymal stem cells,MSCs)对增生性瘢痕成纤维细胞(hypertrophic scar fibroblasts,Fbs)的影响及作用机制。方法分离培养Fbs后,将其分成正常组(N组)和MSCs培养上清组(mesenchymal stem cellconditioned medium,MSCs-CM组)。采用Cell Counting Kit-8(CCK8)和免疫荧光染色检测增殖能力,采用细胞划痕实验检测迁移能力,采用定量实时聚合酶链反应(q RT-PCR)检测转化生长因子β1(Transforming Growth Factor-β1,TGF-β1)基因表达水平,采用酶联免疫分析(ELISA)检测各实验组中白细胞介素10(Interleukin-10,IL-10)表达情况。结果与N组相比,MSCs-CM组Fbs的增殖能力、迁移能力和TGF-β1的基因表达水平均受到抑制(P<0.05),但是两组IL-10的表达水平比较,其在MSCs-CM组中表达明显升高(P<0.05)。结论 MSCs可能通过旁分泌作用释放IL-10对Fbs产生抑制作用。
